Background: The intracellular hyaluronan-binding protein 4 (HABP4 or iHABP4), also designated Ki-1/57, is a cytoplasmic and nuclear protein that is phosphorylated on serine and threonine residues. HABP4 is expressed as a protein of 413 amino acids and is phosphorylated by phorbol 12-myristate 13-acetate (PMA)-activated PKC isoforms at Thr-354 and Thr-375. HABP4 interacts with hyaluron, the adaptor protein RACK1, the transcription factor MEF2C and the chromatin remodeling factor CHD3, suggesting that it may be involved in the regulation of transcription. However, its actual function has yet to be elucidated. HABP4 is highly expressed in brain, heart, and kidney and is moderately expressed in skeletal muscle. Also, HABP4 is expressed in a variety of tumor cell lines and in activated, but not resting, leukocytes.
